About Plant Preservative Mixture (PPM)

PPM™ is a heat stable preservative/biocide which can be used to effectively prevent or reduces microbial contamination in plant tissue culture.

  • At optimum doses, PPM™, which stands for Plant Preservative Mixture™, is an extremely effective Preservative/Biocide, yet does not impair in vitro seed germination, callus proliferation and callus regeneration.
  • PPM™ prevents the germination of both bacteria and fungi spores. It is heat stable and therefore can be autoclaved with the media.
  • PPM™ can be, and should be used as a standard ingredient in plant tissue culture media, and is also substantially less expensive than commonly used antibiotics.
While PPM™ was principally designed to inhibit airborne contamination, waterborne contamination and contamination introduced from human contact, it can also -- in many cases -- be used to reduce endogenous contamination.

Despite the most stringent use of sterile techniques, the contamination of plant cell and plant tissue cultures remain a persistent problem that can result in losses ranging from small number of cultures to the loss of whole batches.

    Much less Contamination

Posted by Bill on Dec 13, 2022

I use 1.5mL of PPM per liter of MS media for Ribes propagation and have much less contamination issues than without. I use it for all now.

    Volume 1 Liter

Posted by Johnson Aritonang on Mar 20, 2021

I buy 1 Liter, but maybe customs officers in Indonesia took 250mL as sample, so I only get 750mL. Well, that's okey. I give 5 stars to seller.

    Positive results in emergency situation

Posted by Laurent L on Jan 28, 2021

I had to unexpectedly flask an orchid green seed capsule with no access to an autoclave, laminar flow cabinet or glove box, I used only rudimentary sterilizing techniques and PPM. No contamination after nearly four weeks. Amazing!

    Amazing

So just to test the ability of the PPM to resist microbial contaminants - I dosed two plates of media, one with and one without, with several drops of actively aerated compost tea. In 7 days the plate without PPM was entirely overrun with fungi and bacteria - the plate with PPM has had zero visible growth at 1ml/l. Every culture I have placed in PPM media - has had no problems with unwanted contamination. Some magic stuff here.
