About Plant Preservative Mixture (PPM)

PPM™ is a heat stable preservative/biocide which can be used to effectively prevent or reduces microbial contamination in plant tissue culture.

  • At optimum doses, PPM™, which stands for Plant Preservative Mixture™, is an extremely effective Preservative/Biocide, yet does not impair in vitro seed germination, callus proliferation and callus regeneration.
  • PPM™ prevents the germination of both bacteria and fungi spores. It is heat stable and therefore can be autoclaved with the media.
  • PPM™ can be, and should be used as a standard ingredient in plant tissue culture media, and is also substantially less expensive than commonly used antibiotics.
While PPM™ was principally designed to inhibit airborne contamination, waterborne contamination and contamination introduced from human contact, it can also -- in many cases -- be used to reduce endogenous contamination.

Despite the most stringent use of sterile techniques, the contamination of plant cell and plant tissue cultures remain a persistent problem that can result in losses ranging from small number of cultures to the loss of whole batches.

    I am very grateful to have found PPM. In the past I was averaging a 70% contamination rate. Since using PPM I'm holding steady at a 5% contamination rate.

    I'm new to micropropagation. I was concerned about contaminated cultures. I've use PPM in the first several media batches and all have been contamination-free. For me, PPM is a required ingredient in a media formulation.

    I am a beginner hobbyist in tissue culture, still experimenting to get reliable results. There is a notable improvement in contamination rates when using PPM. I recommend it and will continue to use it in my culture medium.

    Had fungal growth in my latest batch of water-germinated VFT seeds. After rinsing the seeds w/0.5% bleach and then plating it into MS+PPM, they germinated without any contaminants. PPM works.

    Adding the recommended amount of heat-stable PPM (2 ml per liter of orchid germination medium) provides assurance that small lots of imported orchid seed received under the USDA APHIS 587 program are more likely to be sown successfully without competing, destructive contamination.
