What is Plant Preservative Mixture (PPM) ?

Plant Preservative Mixture (PPM), is a broad-spectrum biocide/ for plant tissue culture. PPM™ is the ultimate solution to the never-ending struggle against microbial airborne, waterborne and endogenous contamination.

About Plant Preservative Mixture (PPM)

PPM™ is a heat stable preservative/biocide which can be used to effectively prevent or reduces microbial contamination in plant tissue culture.

  • At optimum doses, PPM™, which stands for Plant Preservative Mixture™, is an extremely effective Preservative/Biocide, yet does not impair in vitro seed germination, callus proliferation and callus regeneration.
  • PPM™ prevents the germination of both bacteria and fungi spores. It is heat stable and therefore can be autoclaved with the media.
  • PPM™ can be, and should be used as a standard ingredient in plant tissue culture media, and is also substantially less expensive than commonly used antibiotics.
While PPM™ was principally designed to inhibit airborne contamination, waterborne contamination and contamination introduced from human contact, it can also -- in many cases -- be used to reduce endogenous contamination.

Despite the most stringent use of sterile techniques, the contamination of plant cell and plant tissue cultures remain a persistent problem that can result in losses ranging from small number of cultures to the loss of whole batches.

    Mi experiencia con PPM

Posted by Álvaro Araya López on 19th Jun 2020

I have used PPM for over 6 years now with my lilium tissue culture media and I have had very little contamination with it's use in less than .5%.

    The best in the market

Posted by Julian Resurreccion on 19th Jun 2020

I used 4ml on a starting media, and so far i havent gotten any issue. My first try, all my batch grew mold after a week. This time, its been a week and a half and i havent seen any mold in my tissue culture. Also, sjopping around, i find this to be more economical where others are quite pricy. This one does the job and more.would buy again

    PPM efficacy

Posted by clinton boyd on 18th Jun 2020

From my experience, PPM is superior to the conventional use of antibiotics and anti-fungal agents in culture media.
My contamination rate has fallen dramatically to about 1%. Before PPM I was getting up to 60% contamination.

I use PPM for disinfection of the dry orchid seeds before sowing on media containing PPM and I get good germination and very low contamination.

    PPM efficacy

I have been doing orchid seed growing and replating for many years. The primary problem has consistently been contamination- even after stringent disinfection of the seeds with hypochlorite or Hydrogen Peroxide, sterilisation of the culture media in a pressure cooker and aseptic transfer techniques.

I have used antibiotics in combination with anti-fungal agents in my media with only limited success.

Since I discovered PPM in 2016, my contamination rate has fallen drastically!

I clean the seeds and get them free from extraneous matter then soak in PPM solution for at least 12 hour then sow aseptically on pre-sterilized media which was prepared with PPM at a concentration of 1ml per litre.
I now have about a 1% contamination rate.

This is the best antimicrobial agent that I have encountered and I will continue using it.

A powerful game changer when it comes to controlling contamination. Used with and without proper sterilizing techniques vastly reduces the growth of contamination and increases survivability of the clones.

i would recommend this to anyone tissue culturing, as it will increase success rate
