  1. Prepare liquid tissue culture media using PCT’s MS media (4.54 grams) and add 25-30 grams of sugar, 1ml PPM, and 4 grams of PCT gellan gum or 8 grams of PCT agar.
  2. Then, add the desired quantity of the plant hormone from the prepared PGR’s stock solution.
  3. Autoclave the medium for 20 minutes at 15 psi and 121℃.
